Depression Events Associated with Proton-Pump Inhibitors in Postmarketing Drug Surveillance Data

Proton-pump inhibitors, PPIs, are widely prescribed and available over the counter for prolonged reduction of stomach acid production related disorders. PPIs irreversibly inhibit hydrogen/potassium ATPase in gastric parietal cells. Recent retrospective studies have described an association between PPI use depression. However, there is conflicting evidence that therapy improves depressive symptoms. Considering widespread over-the-counter availability these drugs, further investigation into depression adverse event was warranted with a larger-scale postmarketing set reports. Here we analyzed 125,923 reports from FDA Adverse Event Reporting System consisting histamine-2 receptor antagonist monotherapy records found statistically significant Additionally, each six currently marketed individually observed reaction all them.
